Agar gel immuno-diffusion (AGID) tests, historically, have been widely used for the detection of group-specific antibodies against BTV (Pearson et al., 1985) although the development of more rapid, sensitive, specific and higher throughput antibody-detection ELISAhas led to a decline in their use. Older detection systems include the complement fixation assay and tube precipitin antibody test. Complement-fixing type antibodies can be detected in body fluids other than serum, especially CSF which is useful for C. immitis meningitis. Other serological tests on CSF are not as useful. Antibody concentration using complement fixation (CF) is done by testing twofold serial dilutions of patient specimens for complement consumption. Generally, the greater the dilution that still results in complement consumption, the likelihood is higher that the patient has extensive infection. The CF test is based upon the depletion of complement that results after antibodies in a specimen from a patient infected with C. immitis form an immune complex when mixed with coccidiodal antigen. Complete depletion is detected by failure of tanned red blood cells to lyse when added to the mixture because these cells are normally lysed in the presence of complement. The coccidioidal antigen used is a complex extract of C. immitis, but it is also known that chitinase is involved in these reactions which is an important protein enzyme component of the fungal cell wall structure.

The use of VIG has become extremely limited since the eradication of smallpox. It is considered valuable “insurance,” to be held in reserve if a patient is receiving an experimental vaccine that involves a vaccinia carrier virus, or to prevent or manage complications of smallpox vaccinationshould such be required for a bioterrorism threat. Recently, a preparation of VIG suitable for IV use (VIG-IGIV) has gained FDA approval. Phosphatidylethanolamine (PE) is a class of phospholipids found in biological membranes.[1] They are synthesized by the addition of cytidine diphosphate-ethanolamine to diglycerides, releasing cytidine monophosphate. S-Adenosyl methionine can subsequently methylate the amine of phosphatidylethanolamines to yield phosphatidylcholines. It can mainly be found in the inner (cytoplasmic) leaflet of the lipid bilayer.[2]
Phosphatidylethanolamines are found in all living cells, composing 25% of all phospholipids. In human physiology, they are found particularly in nervous tissue such as the white matter of brain, nerves, neural tissue, and in spinal cord, where they make up 45% of all phospholipids.[3]
Phosphatidylethanolamines play a role in membrane fusion and in disassembly of the contractile ring during cytokinesis in cell division.[4] Additionally, it is thought that phosphatidylethanolamine regulates membrane curvature. Phosphatidylethanolamine is an important precursor, substrate, or donor in several biological pathways.[3]
As a polar head group, phosphatidylethanolamine creates a more viscous lipid membrane compared to phosphatidylcholine. For example, the melting temperature of di-oleoyl-phosphatidylethanolamine is -16 °C while the melting temperature of di-oleoyl-phosphatidylcholine is -20 °C. If the lipids had two palmitoyl chains, phosphatidylethanolamine would melt at 63 °C while phosphatidylcholine would melt already at 41 °C.[5]Lower melting temperatures correspond, in a simplistic view, to more fluid membranes.
